Materials Required

  1. Human T cell culture media.
  1. Human cytokines for optimal expansion, such as IL-2, IL-7, IL-15.

Protocol

  1. Wash Human T-Activator CD3/CD28 Beads
    • Resuspend the beads in the tube and vortex or tilt and rotate to ensure proper mixing.
    • Transfer the desired volume of beads to a new tube.
    • Add an equal volume of PBS (containing 1% HSA) for resuspension.
    • Place the tube on a magnet for 1 minute and discard the supernatant.
    • Remove the tube from the magnet and resuspend the beads in the same volume of PBS (containing 1% HSA).
  1. Separate CD3+ T cells
    • Gently resuspend Ficoll-isolated PBMCs in PBS (containing 1% HSA) and adjust the cell density.
    • Add the washed beads to the PBMCs in the appropriate ratio.
    • Rotate the samples and incubate at room temperature.
    • Dilute the mixture of beads and cells with T cell culture media or PBS (containing 1% HSA) and place the tube on a magnet.
    • Remove the supernatant and resuspend the mixture in T cell culture media containing IL-2.
  1. T cell activation and expansion
    • Count the number of T cells daily starting from day 3.
    • Dissociate the beads and cells by gently blowing the cell suspension during the later stages of cell expansion.
    • When CD3+ T cells exceed 1 × 106 cells/mL, dilute the cells with T cell culture media containing IL-2.
    • At the end of the culture (day 9-14), count the cells and remove the beads using magnets.
